Objectives:The aim of this research is to figure out the role of stem cell transcription factors(OCT-4, SOX-2, c-MYC, KLF-4)in ectopic ossification and to better understand the pathogenesis of new bone formation in AS patients.
Methods: Ligament tissues were collected from hip joints from AS patients and control groups who had the operation of hip replacement. The expression levels of OCT-4, SOX-2, c-MYC, KLF-4 were analyzed via real-time polymerase chain reaction (RT-PCR), Western blot (WB) and immunohistochemistry (IHC).
Results: We found that OCT-4, SOX-2 and c-MYC were higher expressed in AS ligament than controls. More interestingly, during the process of inducing primary fibroblast to osteoblast, OCT-4 increased at first several days and then decreased gradually, but c-MYC was always higher expressed than controls. These results suggest that fibroblast may reprogram itself during this process. We also used cytokines to stimulate fibroblasts and found out that OCT-4, SOX-2 and c-MYC can be upregulated by IL-23 alone.
Conclusions: Our results indicated the critical role of the stem cell transcription factors in the process of fibroblast ossification. Furthermore, we put forward a hypothesis that after a long term of inflammatory cytokine stimulation, fibroblast could dedifferentiate to a pluripotent state and then re-differentiate to osteoblast to participate new bone formation in AS.
