Objective: Develop a murine model of AE-IPF by HSV1 infection and explore the mechanisms of AE-IPF. Methods:(1) Establish AE-IPF model: WT and IL-17A-/- mice were divided into 4 groups:BLM+HSV1; BLM+NaCl;NaCl+HSV1;NaCl+NaCl;(2) Role of ERS in the model: 1)Expression of ERS proteins in lung tissues.2)Two groups: BLM+HSV1+TUDCA and BLM+HSV1+NaCl Results: (1)BLM+HSV1 displayed typical features of ALI, and relieved in KO mice. Survival rates of BLM+HSV1(15% 3/20) were lower than that of KO mice(11/20 55%)(P=0.0254).Levels of IL-6(P=0.0188),KC(P<0.0001) and G-CSF(P=0.0045) in BALF were significantly higher in BLM+HSV1 than KO mice. (2) Protein levels of ATF4(P<0.0001), ATF6(P=0.0257), and CHOP (P=0.0005)were elevated in BLM+HSV1 compared with BLM+NaCl. TUDCA-treaded mice had decreased neutrophils(P=0.0358) in BALF and TH17(p=0.0036) in peripheral blood. Conclusions: HSV1 infection on BLM-induced fibrosis successfully establishes a kind of AE-IPF murine model, both IL-17A and ERS may contribute to AE-IPF.
