论文摘要: Background: In this study, we investigated the roles of hsa-circ-0066070, miR-92a and CCND1 in the PAH.
Method: CircRNA realtime PCR to search circRNA involved in PAH, real-time PCR and western-blot anlaysis were performed to detect miR-92a and CCND1 between different group, as well as effect of miR-92a on CCND1, or effect of hsa-circ-0066070 on miR-92a and CCND1. Finally, MTT assay and FCM (Flow cytometry) were performed to detect effect of hsa-circ-0066070 on cell proliferation and cell cycle.Result: MiR-92a level in COPD (+) PAH(+) group was much lower, while CCND1 level in COPD (+) PAH(+) group was much higher than COPD (+) PAH(-) group. While transfecting with hsa-circ-0066070 shRNA up-regulated miR-92a level, and down-regulated CCND1 expression. Hsa-circ-0066070 evidently promoted cell viability, and cell number of cells transfected with hsa-circ-0066070 in G1/G0 phase was less, and in S phase was much greater than control.Conclusion: Hsa-circ-0066070 was associated with PAH via influencing smooth muscle cell proliferation induced by miR-92a/CCND1.
