论文摘要: BACKGROUND Serum response factor (SRF) was previously found to be involved in the phenotypic transition and fibrosis of the peritoneal membrane during treatment with peritoneal dialysis (PD), but the exact mechanism was unclear.
METHODS We showed in this study that miR-199a/214 gene cluster, which has a CArG element in the promoter, was directly regulated by SRF.
RESULTS Expression of miR-199a-5p/214-3p gene cluste was significantly unregulated in human peritoneal mesothelial cells (HPMCs) after HG treatment.  By chromatin immunoprecipitation and reporter assays, we found that SRF could bind to the miR-199a-5p/214-3p gene cluster promoter after HG stimulation.  The silencing of miR-199a-5p or miR-214-3p could inhibit phenotypic transition of HPMC induced by HG, and alleviate HG-induced damage and fibrosis ain PD rats in vivo. In vitro assay suggested that silencing of miR-199a-5p or miR-214-3p promoted migration but inhibit adhesion of HG induced HPMC.  Both miR-199a-5p and miR-214-3p were found to target the claudin-2 (CLDN2) and E-cadherin (CDH1) messenger RNAs.
CONCLUSIONS Overall, this study reveals a novel SRF-miR-199a/miR-214-CDH1/CLDN2 axis that mediates damage and fibrosis in PD.
