Objective: To assess the role of AQP2 in embryo implantation.
Study Design: Normal endometrial samples were collected from women who attended our hospital for IVF-ET. Samples were categorized as diferrent phases. AQP2 mRNA and protein expression in response to E2 was assessed. The effects of AQP2 on spheroid attachment were assessed by an in vitro co-culture assay siRNA against AQP2.
Results: The highest levels of AQP2 were observed during the late proliferative and mid-secretory phases. The lowest were observed during the early proliferative and late secretory phases. Both10-9 and 10-7 M E2 for 24h up-regulated AQP2 in cultured endometrium.Knockdown of AQP2 by siRNA significantly decreased JAR spheroids attachment, with or without E2(P<0.01).
Conclusion: AQP2 expression in human endometrium is mediated by estrogen. Low AQP2 level could be a potential cause for impaired uterine receptivity.
